The present study aimed to develop and evaluate a lidocaine-loaded mucoadhesive film-forming spray for localized periodontal pain management. Lidocaine was characterized by UV spectroscopy, melting-point determination, DSC and FTIR, followed by preformulation and drug–excipient compatibility studies. An optimized formulation containing 2% lidocaine, 2% Eudragit RSPO, 3% HPMC K4M and an isopropyl alcohol ratio of 6:4 was evaluated for film formation, drying time, spray pattern, drug content, mucoadhesive force and in-vitro drug release. The formulation formed a homogeneous film, dried within 60 s, showed a uniform spray pattern and exhibited a spray angle of 82.4°. Drug content was 4 mg per actuation, with a mucoadhesive force of 0.1078 N. Cumulative lidocaine release reached 52.29% at 360 min, compared with 98.23% for the marketed comparator, with the Higuchi model providing the best fit (R² ≈ 0.993). The developed spray demonstrated satisfactory film-forming and mucoadhesive properties with prolonged drug release, indicating its potential as a localized periodontal drug-delivery system.
